These applications use a single-frit PhyTip column design, with the media bed loose in the column above a bottom frit. The design still benefits from the frit-air barrier, offering very high control over fluid flow, and DFC (Dual Flow Chromatography) target sample concentration effects. The advantage is that resin bed can accommodate much higher levels of cell debris and sample variation – an essential requirement to accommodate full automation. PhyNexus’ proprietary modified chaotropic silica chemistry fully loads the media in 5-10 flow cycles (depending on sample concentration levels) and elutes extremely low endotoxin DNA without requiring additional wash cycles.
Plasmid Purification PhyTip Column and Bed Sizes
PhyNexus offers two primary PhyTip Column sizes with varying resin bed sizes depending on the prep scale desired: 1000 μL columns for Miniprep most major 96-channel instruments,, 1000 μL columns for Midiprep compatible with the PhyNexus AutoPlasmid MEA instrument, and 20 mL columns for Maxi, Mega and Gigaprep scale compatible with the PhyNexus AutoPlasmid MMG instrument.
