BAC Capture Select PhyTip Columns

BAC CaptureSelect® products are affinity ligands created by a proprietary technology based on Camelid-derived single domain antibody fragments. The ligand is a 12-kD single domain fragment which comprises the 3 CDRs that form the antigen binding domain, efficiently produced by the yeast Saccharomyces cerevisiae.

Advantages of CaptureSelect® Resins

  • Tunable Specificity The specificity of the ligand can range from broad to very narrow: species, antibody class, subclass format, glycoform, isomer, idiotype, etc. Specific features can be incorporated into the screening process such as binding and elution conditions. The screening for selectivity and process functionality has proven to be highly effective in identifying ligands that meet customer requirements regarding defined purification processes.
  • Optimal Affinity The binding affinity is in the nmol range and can be optimized for both capture and scavenging applications. This affinity enables capture of valuable biotherapeutics at very low concentrations.
  • Excellent Quality Since the DNA that codes for each ligand is amplified by PCR and subsequently cloned and expressed in bakers yeast, all ligands are non-animal derived. This elegant means of production results in superior products with excellent and consistent quality.
Description

a) CaptureSelect LC-kappa (Hu)

This affinity matrix is specifically designed to purify all human kappa Ig light chain (subclass I, II, III and IV) containing antibody formats and fragments, including kappa Fab, kappa Fab2′, and all subclasses of human kappa IgG, IgA, IgD, IgE, and IgM. The matrix is based on a proprietary ligand that recognizes human kappa light chains through high affinity binding at the light chain constant domain. Unlike protein L which binds the variable domain and ignores a significant fraction of the kappa light chain population, this product binds 100% of human kappa light chains. In contrast to protein L this product has a high specificity for human kappa light chains, providing the ability to separate human kappa light chain populations from transgenic expression systems. Furthermore, this product can also be used to elute under gentle elution conditions.

b) CaptureSelect LC-lambda (Hu)

This affinity matrix is specifically designed to purify all human lambda Ig light chain (subclass I, II, III and IV) containing antibody formats and fragments, including lambda Fab, lambda Fab2′, and all subclasses of human lambda IgG, IgA, IgD, IgE, and IgM. The matrix is based on a proprietary ligand that recognizes human lambda light chains through high affinity binding at the light chain constant domain. This product has a high specificity for human lambda light chains, providing the ability to separate human lambda light chain populations from transgenic expression systems. This product can also be used to elute under gentle elution conditions.

c) CaptureSelect IgG-Fc (Hu)

This affinity matrix is specifically designed to purify all human IgGs (including IgG3) and Fc fusion proteins from cell culture and transgenic expression systems, as well as from human serum and plasma. The matrix is based on a proprietary ligand that recognizes all four subclasses of human IgG through high affinity binding at the Fc region of the heavy chain. Unlike Protein A this product has a high specificity for human IgGs, providing the ability to separate human IgG populations (both monoclonal and polyclonal) from transgenic expression systems. In addition, in contrast to Protein A this product also can purify human IgG3 along with human IgG 1, 2 and 4, and it can be used to purify polyclonal human IVIG from plasma and serum.

d) CaptureSelect IgG-Fc (ms)

This affinity matrix is specifically designed to purify IgGs from multiple species including mouse, cow, rabbit, rat, goat, horse, human and sheep. For a complete list of species refer to the product sheet. The matrix is based on a proprietary ligand that recognizes IgGs from multiple species through high affinity binding at a highly conserved epitope on the Fc region of the heavy chain. Unlike Protein, G, which, under standard elution conditions, is known to present challenges that result in poor yield, dissociated homodimers, and denatured antibodies, this product can be used with gentle elution conditions, retaining the structural and functional integrity of the eluate.

e) CaptureSelect IgM

The CaptureSelect IgM affinity matrix contains a ligand recognizing a unique domain on the Fc part of IgM that enables purification of human and mouse IgMs without cross reactivity with human and mouse IgG and IgA. This makes the CaptureSelect IgM affinity matrix a superior resin in comparison with other IgM matrices in terms of specificity and product purity. The ligand is coupled to NHS-activated Sepharose 4 Fast Flow® (GE Healthcare). This matrix can be used for the purification and isolation of IgM, without cross-binding to other immunoglobulins, from complex sources like plasma, serum, and cell culture supernatants

Application

PhyTip columns are unique capture, purification and enrichmentTM tools from PhyNexus designed for micro volume protein sample preparation. PhyTip Columns are available for a variety of liquid handling platforms and contain specific affinity resins for application specific requirements.

Data

PhyNexus recommends the use of the PhyTip Starter Kit containing IgG as a standard to verify the use of PhyTip columns with all applications.

Samples for purification and enrichment must be clear and free from particulate matter. It is highly recommended to centrifuge samples and use the clear supernatant only, prior to use with PhyTip columns.

Shipping and Storage

Each pack of PhyTip columns has been manufactured and QC’d to the highest standards and shipped in retainer boxes that maintain the integrity of the specific affinity resin within each PhyTip column. This product is shipped at ambient temperatures, but on receipt should be stored in a standard laboratory refrigerator between 4 and 8◦C.

  • Do NOT freeze or store frozen.
  • When not in use, keep the lid of the box closed and sealed, store in the refrigerator.
  • Do not allow affinity resin to dry out by extended storage in a dry environment.

PhyTip columns with Protein G are stored in Glycerol when shipped from PhyNexus.
