Mass Spec Sample Prep and the Need for an Automated Workflow
Mass spectrometry is a powerful tool for qualitative and quantitative analysis of peptides, proteins and protein complexes, and an increasingly important part of drug discovery and development. MS can accurately determine the mass and characterization of proteins, provide antibody mapping and peptide mapping for biomarker discovery, identify post-translational modifications, map protein complexes and give insight to protein-protein and protein-to-lipid interactions. As such, labs are analyzing ever more analytes across multiple screening conditions.
But sample prep for MS can be slow and labor intensive, involving analyte capture, purification and enrichment, and subsequent analyte clean-up of salts and surfactants – which are incompatible with MS – with C18 resins.

PhyNexus C18 PhyTip Columns offer an automated solution, compatible with existing liquid handling robotics, to sample clean-up for MS. Moreover, as part of LC-MS workflow, C18 PhyTip Columns can be integrated with upstream sample purification and concentration steps on an HTP automated system to vastly reduce labor, increase throughput and allow for significant conditions screening variability.
PhyNexus C18 columns bind proteins and peptides through reverse phases solid phase extraction (SPE). The C18 PhyTip Columns contain one of two resin options between two inert screens at the column tip. The liquid handler (handheld, bench top, or HTP 96-channel) then processes the sample with the back and forth flow of Dual Flow Chromatography to drive the binding reaction to saturation. C18 PhyTip Columns deliver excellent results with all the benefits of automation.
